Description: λ DNA is used as one of the substrates in restriction enzymes research and for testing of restriction endonucleases activity. The double stranded DNA is isolated from bacteriophage lambda (cl857 ind1 Sam7). The molecular weight is 31.5 x 10⁶ daltons and it is 48,502 base pairs in length.

Preparation: The phage is isolated from the heat inducible lysogen E. coli λ cl857 S7 by gel filtration. The DNA is isolated from the purified phage by phenol/chloroform extraction and dialyzed against 10 mM Tris-HCl (pH 7.4) and 1 mM EDTA.

Quality control: Gel analysis for purity. EcoRI and HindIII fragmentation patterns.

Storage buffer: 10 mM Tris-HCl (pH 7.4), 1 mM EDTA. Store at -20^oC.
